Earlier this season, there was speculation surrounding Mainoo's future at Old Trafford after he was out of favour under former United head coach Ruben Amorim.
However, since Amorim left the club in January, the midfielder has returned to the first-team fold under interim boss Michael Carrick and played a key role in helping United to the brink of playing UEFA Champions League football next season.
"Manchester United has always been my home; this special club means everything to my family," Mainoo said in a statement on the club's website.

"I have grown up seeing the impact that our club has on our city, and I relish the responsibility that comes with wearing this shirt.
"From The Cliff, to Littleton Road, to Carrington and ultimately Old Trafford, the journey so far has been incredible. I have the privilege of living my dream every single day, with the same relentless desire to succeed here as when I joined my first training session at the age of six.
"We can all feel the momentum building inside the club. I am determined to step up and play my role in helping Manchester United to regularly fight for major trophies in the years ahead."

The 21-year-old has been a mainstay in the Manchester United first team since Carrick's return to the Old Trafford helm back in January and his form under the former Middlesbrough boss saw the midfielder return to the England setup last month.
Mainoo played both games as England were held to a 1-1 draw with Uruguay and crashed to a 1-0 defeat to Japan at Wembley during the March international break.
The youngster follows in Harry Maguire's footsteps, the centre-back having put pen to paper on a new contract until 2027, with the option for a further year, at the start of April.
Like Mainoo, Maguire also earned an England recall in March as reward for his form under Carrick, with the defender putting in an assured display in the stalemate with South American powerhouse Uruguay.

Manchester United will hope to follow up their 2-1 win over Brentford on Monday night with another three points when they welcome rivals Liverpool to Old Trafford on Sunday.
Victory over The Bees leaves Manchester United 11 points clear of sixth-placed Brighton with four games to go.
Just three points, meanwhile, separate Manchester United and Liverpool before the weekend's clash with The Reds seeking to end a testing season on a high.

Arne Slot has overseen three successive victories over Fulham, Everton and Crystal Palace, and a win over United will all-but secure UEFA Champions League football next season.
Liverpool are unbeaten in their past two league visits to Old Trafford having followed up a 2-2 draw in Jurgen Klopp's final game at the ground with a resounding 3-0 victory in Slot's first match against their great rivals during his debut season at the Anfield helm.
